Polyhydroxyalkanoates (PHA) represent a promising class of biodegradable polymers that can serve as sustainable alternatives to conventional petroleum-based plastics. However, their broader industrial application is often limited by insufficient material properties, particularly the high crystallinity and brittleness of poly(3-hydroxybutyrate). One effective strategy to overcome these limitations involves the production of PHA heteropolymers, enabling tuning of their physicochemical and mechanical properties. Thermophilic bacteria of the genus Aneurinibacillus have recently emerged as promising yet still underexplored microbial cell factories. Their metabolic versatility allows the incorporation of structurally diverse monomers into PHA chains without the need of genetic modification. This work focuses on expanding the understanding of substrate utilization and polymer biosynthesis in thermophilic Aneurinibacillus strains, with emphasis on the production of functionalized and structurally diverse PHA. A broad spectrum of precursor compounds, lactones and diols, was evaluated. Lactone-based precursors were generally efficiently utilized, leading to significant incorporation of monomers such as 4-hydroxybutyrate, 5-hydroxyvalerate, and 4-hydroxyhexanoate. In contrast, incorporation of diol-based precursors showed a strong dependence on cultivation conditions and were effectively utilized mainly in defined mineral salt media. Additionally, PHA containing structurally unique α-methylated units derived from α-methyl-γ-butyrolactone were successfully produced and characterized using FTIR, DSC, and TGA. PHA polymers were further modified using terminal alkyne-containing alcohols, enabling the introduction of reactive functional groups. The incorporation of alkyne functionalities was confirmed by ¹H NMR, and the polymers were characterized by SEC-MALS. Their ability to undergo click reactions was subsequently verified using SEC-MALS with UV detection, confirming successful post-polymerization modification. Beyond PHA production, Aneurinibacillus spp. also exhibited extracellular enzymatic activities and biosurfactant production, as demonstrated by emulsification activity, surface tension reduction, and anthracene solubilization. Overall, this work highlights the versatility of thermophilic Aneurinibacillus spp. as producers of tailor-made biodegradable polymers and other value-added products, supporting their potential in sustainable biotechnological applications.
