This thesis focuses on the determination of residues of selected pharmaceuticals, specifically beta-blockers and nonsteroidal anti-inflammatory drugs (NSAIDs), in plant matrices, with a focus on common spinach (Spinacia oleracea L.). The theoretical section summarizes the basic characteristics of the drugs, including relevant properties such as their mechanism of action and the main sources of their release into the environment. It further examines their fate, both in terms of biotic and abiotic processes, and assesses their potential toxicological effects. The experimental section focuses on studying the uptake and accumulation of selected drugs in the aboveground parts of plants using hydroponic and soil experiments with common spinach. As part of this work, the QuEChERS extraction method was optimized for this plant matrix with the aim of achieving the highest possible extraction efficiency and reproducibility of results. Liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) was used for the subsequent analysis of the samples The results showed that the degree of degradation and accumulation of pharmaceuticals strongly depended on the physicochemical properties of the individual compounds and the type of environment. Ibuprofen exhibited the highest degradation rate, whereas atenolol, nadolol, meloxicam, and piroxicam proved to be relatively persistent compounds. The highest accumulation in spinach was observed particularly for atenolol and propranolol, whereas ibuprofen and naproxen showed low uptake by plants. At the same time, it was found that lower concentrations of pharmaceuticals could stimulate spinach growth, whereas higher concentrations led to growth inhibition and reduced biomass production, especially in the root system. The results further suggest that some pharmaceuticals may be transferred into edible plant tissues and could therefore potentially enter the food chain

Studentka při obhajobě prezentovala náplň, výsledky a závěry své diplomové práce. Následovalo čtení posudků vedoucí a oponentky a byly zodpovězeny položené otázky. Při diskuzi k tématu diplomové práce se zapojili i další členové komise se svými otázkami: prof. Ing. J. Krajčovič, Ph.D. – Sledovala jste i metabolity léčiv? doc. Ing. T. Rozsypal, Ph.D. – Dají se Vaše výsledy extrapolovat na reálné podmínky? doc. Ing. P. Krystyník, Ph.D. – Čím by mohla být způsobena výtěžnost 140 %? Diplomatka odpověděla na všechny doplňující otázky členů komise, které byly v průběhu diskuze k dané problematice vzneseny. Předvedla, že má kvalitní přehled v řešené problematice, a to jak z teoretického, tak praktického hlediska. Po diskusi následovalo hodnocení závěrečné práce. Diplomantka prokázala velmi dobrou orientaci v dané problematice i schopnost samostatné prezentace dosažených výsledků.
