Projekt je zaměřen na studium úlohy membránových přenašečů (efluxových pump) v toleranci k rozpouštědlům u Gram pozitivních bakterií na příkladu butanolu a rozpouštědlotvorných klostridií. Geny potenciálních efluxových pump budou vybrány na základě bioinformatických analýz genomů sekvenovaných klostridií společně s RNA-seq a RT-qPCR transkriptomovými analýzami. Tyto geny budou využity pro získání mutantů nesoucích buď „over-expresi“ nebo „knock-out“. Získané mutanty budou testovány na změnu tolerance k butanolu a změny v transportu látek přes buněčnou membránu pomocí fluorescenčních technik, fluorimetrie a průtokové cytometrie. Kromě cílené mutageneze bude pro získání mutantů využita také technika laboratorní adaptivní evoluce, kdy bude eflux indukován nebo potlačen pomocí přídavku induktorů nebo inhibitorů efluxu. Data získaná pomocí RNA-seq techniky budou využita pro modelování signálních drah zodpovědných za eflux a toleranci k butanolu. Výsledky projektu přispějí k objasnění procesu exkrece rozpouštědel u Gram pozitivních bakterií.

Description in English
This project is focused on the role of membrane transporters (efflux pumps) in solvent tolerance of Gram positive bacteria using, as an example, butanol and solvent-producing clostridia. Based on bioinformatic analyses of potential efflux pumps in sequenced clostridia, and RNA-seq plus RT-qPCR transcriptome analyses, candidate (efflux) genes will be identified, and over-expression or knock-out mutants will be produced. These mutants will be tested for butanol tolerance, and changes in membrane transport will be assessed by the fluorescence-based methods, fluorimetry and flow cytometry. In addition to targeted mutagenesis, we will carry out tolerance studies using adaptive laboratory evolution experiments with efflux inducers and inhibitors. RNA-seq data will be used to model signaling pathways responsible for butanol efflux and resistance. Project results will increase our understanding of solvent excretion in Gram positive bacteria.
