This thesis investigates the operating principles of the FluorCam imaging systems developed by Photon Systems Instruments and the physical and biological background of chlorophyll fluorescence in plants. Its main goal is to develop a method for automatically evaluating the spatial homogeneity of the excitation illumination, supported by a dedicated measurement workstation that integrates the mechanical construction, control electronics, and software for automated data acquisition and processing. Based on the acquired data, two approaches to correcting the identified inhomogeneity are proposed and compared, both leading to improved optical parameters of the system, particularly in terms of light field uniformity. An optimal solution is identified from this comparison with respect to the achievable correction accuracy, implementation complexity, and economic efficiency. The final part presents a survey of currently available high-power LED emitters suitable for excitation illumination in fluorescence imaging systems and compares them with the components used in existing FluorCam devices. The outcome is a set of recommendations for the modernization of the illumination subsystem in serial production.

Student seznámil státní zkušební komisi s řešením své diplomové práce a zodpověděl otázky a připomínky oponenta. Dále odpověděl na otázky komise: Zohlednil jste spektrum vlnových délek osvětlení? Student pohotově odpovídal, trocha se zasekl na bílém světle, ale potom se nedorozumění vysvětlilo a student bez problému vysvětloval dále. S komisí komunikoval o bílých LED a důvodech, proč nepoužil modrou diodu. Co by v práci optimalizované a o kolik se to zlepšilo? Student vysvětlil, že optimalizované bylo osvětlení pomocí filtrů.
