This thesis deals with the detection of biomolecules using electrochemical sensors modified with graphene derivatives. It primarily focuses on the selective detection of a key neurotransmitter, dopamine. The theoretical part comprises a literature review focusing on graphene derivatives, electrochemical sensors, their classification, properties, and applications in the analysis of biologically significant compounds. The experimental part is devoted to the development of a sensor based on a screen-printed carbon electrode (SPCE), the surface of which was sequentially modified with undoped graphene acid (GA), nitrogen-doped graphene acid (GAN), and a Nafion polymer layer. Cyclic voltammetry and electrochemical impedance spectroscopy were utilized for the detailed characterization of surface modifications and the electrochemical properties of the phase interface. The analytical parameters of the developed sensors were subsequently evaluated using constant-potential amperometry. Investigation of the modification effects demonstrated that the system combining the GAN material with an optimized 0.25% Nafion layer (SPCE/GAN/Nafion) is the most suitable for the reliable detection of dopamine. The developed sensor is characterized by high sensitivity and, owing to the permselective nature of the polymer membrane, exhibits absolute selectivity coupled with the complete suppression of the electrochemical response of interfering ascorbic acid.

Studentka obhájila bakalářskou práci a odpověděla na otázky členů komise a oponenta.
