This bachelor's thesis focuses on the validation and evaluation of a new version of the PerfLab software tool, developed at the Institute of Scientific Instruments of the Czech Academy of Sciences, for the perfusion analysis of DCE-MRI data. The aim of the work was to propose evaluation metrics and verify the accuracy and repeatability of the arterial input function and perfusion parameter estimation. The analysis was conducted on both synthetic and clinical datasets from the international OSIPI-DCE challenge, enabling a direct comparison of the newly computed results with the outcomes of the original version of the PerfLab software. Within the methodological evaluation, various approaches for determining the shape of the arterial input function were tested, including the dominant reference tissue method for its scaling, as well as the influence of pharmacokinetic models and optimization algorithms on the estimation of perfusion parameters. The results demonstrated that blind deconvolution methods represent a stable solution for clinical practice. For the calculation itself, the regularized optimization algorithm proved to be the most reliable, exhibiting high stability against noise and providing highly reproducible results. Compared to the older version, the new version of the PerfLab software achieves higher reliability and accuracy of estimations. By directly verifying the software's functionality, this work contributes to the preparation of the PerfLab system for clinical and research deployment.

Dále se doptal na vybraný boxplot v prezentaci. Studentka obhájila bakalářskou práci a odpověděla na otázky členů komise a oponenta.
