This bachelor’s thesis focuses on the classification of genetic variants according to ACMG/AMP recommendations. Genetic variant interpretation represents an important part of molecular genetic diagnostics and plays a key role in assessing the clinical significance of identified variants. Due to the growing amount of sequencing data, there is an increasing need for efficient and standardized approaches to variant classification. The thesis is focused on the design and implementation of a software tool for automated evaluation of selected ACMG criteria and subsequent classification of genetic variants. The implementation includes processing of annotated variants and integration of external databases for automated evaluation of ACMG criteria. The software was evaluated using data from a Slovak cardiomyopathy cohort published by Nagyova et al. and datasets obtained from the University Hospital Ostrava. The impact of different variant calling tools on the resulting variant classification was also assessed. The obtained classifications were subsequently compared with the InterVar tool and the ClinVar database. The achieved results are summarized and discussed.

Studentka prezentovala výsledky své práce a komise byla seznámena s posudky. Ing. Šabatová položila otázku zda databáze slouží pro uživatele bez znalostí bioinformatiky. Jak vypadá Excel soubor propojený s databází a jaký je důvod převedení dat do tohoto prostředí. Kolik je celkem zavedených pravidel v AMP systému. Ing. Smíšek položil otázku na data, která uvidí lékaři při práci s Excel tabulkou. Studentka obhájila bakalářskou práci a odpověděla na otázky členů komise a oponenta.
