Quantitative Phase Imaging (QPI), as a group of label-free imaging techniques, provides valuable insight into many biological processes. Long-term live-cell experiments require stable environmental conditions to preserve natural cell behaviour and ensure reliable results. This work presents the design and implementation of an incubation system for a Linnik interferometer-based QPI microscope developed by the Nanoscopy Group at UiT The Arctic University of Norway. The specific requirements of this microscope motivated the development of a custom incubation system enabling control of temperature, humidity, and gas composition. The implemented solution was validated through mechanical and biological testing, demonstrating stable environmental conditions, and further verified through long-term monitoring of HeLa cells exposed to mild hyperthermia. The results confirmed the suitability of the proposed incubation system for reliable long-term live-cell imaging and biological research applications.

Po otázkách oponenta bylo dále diskutováno: Vysychání vzorku. Vliv vlhkosti na optické komponenty mikroskopu. Dá se nějakou metodou oddělit tloušťka a index lomu vzorku? Důvod ohřevu celého okolí mikroskopu. Studentka na otázky odpověděla.
