This bachelor thesis deals with the study of the production of biologically active compounds by the fungus Lentinula edodes during cultivation on different types of substrates. Bran, buckwheat and pearl barley were selected as cultivation substrates. The aim of the thesis was to evaluate the effect of the substrate used on the chemical composition of the material colonized by mycelium and on the content of selected bioactive compounds. The theoretical part focuses on the description of the biological properties of Lentinula edodes, its cultivation possibilities, chemical composition and biological effects. Attention is paid mainly to polysaccharides, β-glucans, phenolic compounds, flavonoids, proteins, lipids, vitamins and minerals. Furthermore, the antioxidant, immunomodulatory and antimicrobial potential of this fungus is described. In the experimental part, the mycelium of Lentinula edodes was cultivated on three selected substrates. Subsequently, total proteins, phenolic compounds, neutral and reducing carbohydrates, flavonoids, total antioxidant activity, selected lipophilic metabolites and glucans were determined in the samples. The thesis also included an indicative determination of the antimicrobial activity of the extracts against selected microorganisms. The obtained results showed that the individual substrates affected the composition of the colonized material in different ways. Bran appeared to be suitable mainly in terms of the content of phenolic compounds, flavonoids and ubiquinone, while pearl barley primarily supported mycelial growth, as well as the content of proteins, reducing carbohydrates and ergosterol. Buckwheat was of interest mainly in terms of antioxidant activity. The results of the antimicrobial tests were not unequivocal, but they indicated a possible antimicrobial potential of some colonized substrates. The obtained results confirm the importance of substrate selection in the cultivation of Lentinula edodes and may be used in further evaluation of colonized substrates as a source of biologically active compounds.

Obhajoba proběhla podle následujícího schématu: prezentace studentky-vyjádření vedoucí/ho-oponentský posudek-reakce na posudek-diskuse s komisí. Studentka přednesla dobrý výtah výsledků své bakalářské práce, v odpovědích na dotazy oponentské i členů komise nebo v reakcích na připomínky k bakalářské práci projevovala menší neznalosti. V diskusi tak studentka prokázala dobrou schopnost orientace v teoretických i praktických základech problematiky bakalářské práce. Komise, s uvážením hodnocení v posudku a vyjádření vedoucí/ho, zhodnotila její bakalářskou práci celkově jako dobrou. Pekař: Vyrostly by houby i s plodnicemi? Přepočítávala se koncentrace látek na sušinu? Obruča: Jaký byl účel výsledného produktu? Dzik: Jak se dělal resazurinový test?
