The bachelor thesis deals with mixed cultivation of bacterial and yeast probiotics on various types of prebiotic substrates, evaluation of their growth, metabolic, antioxidant and antimicrobial activity and subsequent evaluation of their possible use in cosmetics. In the theoretical review, probiotic bacteria and yeasts and some selected strains are characterized. Similarly, characterization of prebiotics and postbiotics and their use in cosmetics are also described. In the experimental part, nine yeast strains were cultivated: Rhodotorula kratochvilovae, Rhodotorula mucilaginosa, Rhodotorula toruloides, Saccharomyces boulardii, Saccharomyces cerevisiae, Yarrowia lipolytica, Metschnikowia andauensis, Metschnikowia chrysoperlae, Metschnikowia pulcherrima on various media. Their lyophilized biomass was used to prepare two types of yeast extracts: aqueous and combined (water/ethanol). These extracts were used to cultivate two selected probiotic bacteria (Lactobacillus acidophillus and Bifidobacterium bifidum). At the same time, bacteria were also cultivated in various combinations with yeast extracts, MRS medium or inulin. Bacterial growth was evaluated as number of viable colonies (CFU). Prepared extracts and supernatants obtained from individual cultivations were evaluated using spectrophotometric methods for determination of antioxidant activity using the ABTS method and the content of total carbohydrates using the Dubois method. Lactic acid production was determined using HPLC analysis. Furthermore, the disk diffusion method was used to determine antimicrobial activity. Based on the results obtained in this thesis, a possible cosmetic product was designed. The obtained results show that the tested yeast extracts can serve as prebiotic substrates that contribute to increased bacterial growth and metabolite production.

Obhajoba proběhla podle následujícího schématu: prezentace studentky-vyjádření vedoucí/ho-oponentský posudek-reakce na posudek-diskuse s komisí. Studentka přednesla výborný výtah výsledků své bakalářské práce, řádně zodpověděla všechny dotazy oponentské i členů komise, pohotově reagovala na připomínky. V diskusi tak studentka prokázala výbornou schopnost orientace v teoretických i praktických základech problematiky bakalářské práce. Komise zhodnotila její bakalářskou práci celkově jako výbornou. Pekař: Co jsou post-biotika? Jak by měl vypadat navrhovaný produkt? Obruča: Jaký je postup při kultivaci kvasinek a následně bakterií? Používají se už takovéto postupy někde? Jak byly získávány extrakty z kvasinek? Neměly extrakty charakteristický zápach?
