The subject of this bachelor's thesis is the study of the effects of low-temperature atmospheric pressure plasma on microbial biofilms of Escherichia coli and Staphylococcus epidermidis. The work focuses on the evaluation of its decontamination efficiency and biological effects related to cellular activity, including potential applications in wound treatment. A unipolar microwave plasma jet was used for the experiments. The movement of the plasma jet over the sample surface was controlled using computer software. The applied power ranged from 12–13 W. Argon with a purity of 4.6 was used as the carrier gas, with a flow rate set to 5 L/min. The biofilms were treated using the plasma jet, and their viability was subsequently evaluated in comparison with untreated samples. The main monitored parameter was the treatment time of microbial samples using plasma. The decontamination effect was evaluated based on the reduction in colony count. In addition, cell viability was assessed using the resazurin assay, which confirmed decreased metabolic activity after treatment. The results showed that increasing treatment time led to a gradual decrease in the viability of microorganisms. This trend was more pronounced in Escherichia coli, which may be related to differences in cell wall structure or to a higher susceptibility to oxidative stress compared to Staphylococcus epidermidis. For both bacterial species, sample rinsing was also performed, which contributed to increased treatment efficacy. In addition, the influence of the plasma gas and possible sample dehydration was evaluated, as these factors may have contributed to the overall effect of plasma treatment.

Obhajoba proběhla podle následujícího schématu: prezentace studentky-vyjádření vedoucí/ho-oponentský posudek-reakce na posudek-diskuse s komisí. Studentka přednesla výborný výtah výsledků své bakalářské práce, řádně zodpověděla všechny dotazy oponentské i členů komise, pohotově reagovala na připomínky. V diskusi tak studentka prokázala výbornou schopnost orientace v teoretických i praktických základech problematiky bakalářské práce. Komise zhodnotila její bakalářskou práci celkově jako výbornou. Pekař: Jak byl realizován experiment s ofukem argonu? Jak se kontrolovala tvorba biofilmu? Dzik: Jak byla realizována metodika počítání kolonií a následně resazurinového testu?
