This master’s thesis deals with the assessment of the direct and indirect effects of selected pharmaceuticals, specifically oxytetracycline, diclofenac and ketoprofen, and their mixtures on representatives of the aquatic ecosystem, the crustacean Daphnia magna and duckweed Lemna minor. The theoretical part summarizes knowledge on the occurrence, fate and ecotoxicity of these substances in the aquatic environment. The experimental part evaluates and compares the effects of individual pharmaceuticals and their mixtures in standard medium and after passage through the soil matrix, i.e. in soil percolate. This approach made it possible to assess the influence of soil on the bioavailability and toxicity of the studied substances. In D. magna, mortality, body length and reproduction were monitored, while in L. minor, growth parameters, chlorosis and the content of photosynthetic pigments were evaluated. The obtained results showed that the assessment of D. magna mortality or merely the number of duckweed fronds is insufficient for determining toxicity. Sublethal parameters, especially reproduction, root morphology and pigment content, showed more pronounced changes even at low concentrations of some pharmaceuticals. Passage through the soil matrix partially reduced growth inhibition in L. minor and changed the response in D. magna in some tested variants; however, specific morphological and physiological changes were induced in both organisms. The thesis confirms the need to test mixtures and to monitor endpoints not only at the organismal level for an objective assessment of the risks of pharmaceuticals in the environment.

Studentka v rámci obhajoby diplomové práce prezentovala náplň, získané výsledky a závěry práce. Dále byly přečteny posudky vedoucí a oponentky a byly zodpovězeny otázky oponentky. V následné diskusi byly položeny dotazy: doc. Ing. T. Rozsypal, Ph.D. – Můžete popsat výsledky z pohledu významu platných číslic? Diplomatka odpověděla doplňující otázky členů komise, které byly v průběhu diskuze k dané problematice vzneseny. V diskuzi studentka prokázala velmi dobrou orientaci v dané problematice. Po diskusi následovalo hodnocení závěrečné práce. Diplomantka prokázala velmi dobré odborné znalosti z oblasti týkající se diplomové práce.
