This thesis focuses on the study of interactions between hyaluronan (HA) and the TSG-6 protein and serum proteins as a function of environmental parameters. Understanding the mechanism of these interactions, particularly the influence on the formation of HA complexes with heavy chains (HC), could significantly contribute to elucidating their function in inflammatory processes, which could lead to potential future applications in new diagnostic and therapeutic methods. The study was conducted using a quartz crystal microbalance with impedance measurement (QCM-I) method. The experiments evaluated the influence of HA molecular weight and physicochemical environmental factors, specifically temperature, pH, and the presence of magnesium ions. Based on the results obtained, it was demonstrated that environmental factors simulating an inflammatory state (particularly a slightly acidic pH and physiological temperature) significantly increase the binding affinity of the TSG-6 protein to hyaluronan. Furthermore, a marked interactions dependence on the HA chain length was observed. While binding results differed between whole serum proteins and purified inter-alpha-inhibitor (IαI) when tested with medium-molecular-weight HA, similar binding levels were observed with short HA oligosaccharides. At the same time, short oligosaccharides appeared to be the most effective for TSG-6 binding in the experiments, making them promising candidates for future research focused on the issue of irreversible heavy chain (HC) dissociation in the inflammatory environment.

Obhajoba proběhla podle následujícího schématu: prezentace studentky-vyjádření vedoucí/ho-oponentský posudek-reakce na posudek-diskuse s komisí. Studentka přednesla výborný výtah výsledků své diplomové práce, řádně zodpověděla všechny dotazy oponentské i členů komise, pohotově reagovala na připomínky. V diskusi tak studentka prokázala výbornou schopnost orientace v teoretických i praktických základech problematiky diplomové práce. Komise zhodnotila její diplomovou práci celkově jako výbornou. Pekař: Jak si vysvětlujete vliv molekulové hmotnosti na interakci hyaluronanu? Obruča: Vazba čeho na hyaluronan se řešila? Kozáková: S jakou přesností byla stanovována hmotnost? Jakou funkci má v daném systému zlato?
