This diploma thesis deals with the production and characterization of bioactive metabolites of the yeast Metschnikowia pulcherrima. The theoretical part summarizes current knowledge on the taxonomy, cytology, and production capabilities of this species, with a focus on the pigment pulcherrimin and exopolysaccharides (EPS). The aim of the practical part was to optimize the methods for pigment isolation from cell biomass and the extracellular space, perform a comparative analysis of methods for iron determination, and test the prebiotic potential of the obtained EPS. As part of the experiments, an optimized extraction protocol was developed, revealing that pretreatment of the biomass with 89.8% methanol is essential for obtaining a pure isolate with a high yield of pulcherrimin. Analytical comparison of the methods demonstrated that complete microwave mineralization followed by ICP-OES measurement is required for accurate quantification of iron in the stable pulcherrimin complex. Conventional colorimetry showed up to a fifty-fold underestimation of the results due to insufficient sample decomposition. The highest specific iron content achieved in the isolate was 12.30 mg/g, confirming the successful concentration of the pigment compared to the initial biomass (0.49 mg/g). The study of biological properties demonstrated a strong, yet strain-specific prebiotic effect of the isolated EPS. Viability results confirmed that the addition of EPS significantly supports the growth of probiotic bacteria, with the highest stimulation (an increase of up to 1368%) recorded for the strain Lactobacillus casei, while Lactobacillus plantarum also showed excellent results. Furthermore, the thesis provides data on the fatty acid profile, which was dominated by monounsaturated fatty acids (MUFAs), particularly oleic acid, across all cultivations. Quantification of ergosterol and ubiquinone revealed significant differences in their specific production depending on the chosen cultivation scale (flask vs. fermenter). The findings contribute to a better understanding of metabolic processes and expand the possibilities for utilizing M. pulcherrima yeast in industrial applications.

Po diskusi následovalo hodnocení závěrečné práce. Diplomantka prokázala nejen výborné odborné znalosti, ale i schopnost samostatné prezentace dosažených výsledků.
