This bachelor’s thesis focuses on determining the stress resistance of selected strains of the thermophilic bacterium Caldimonas thermodepolymerans to specific stressors. The theoretical section provides a general characterization of polyhydroxyalkanoates (PHA) with a focus on thermophilic producers. At the same time, part of the literature review is devoted to the stress response of cells when exposed to various types of abnormal environments. The experimental part deals with the assessment of the stress resistance of selected strains under acute stress applied at various intensities. To assess the stress role of PHA in the stress response of the thermophilic bacterium Caldimonas thermodepolymerans, two mutant strains with deletions in the genes for PHA-synthase (phaC) and PHA-depolymerase (phaZ) were selected for comparison with the wild-type strain. As a stress factor was chosen osmotic stress which was applied to the strains using sodium chloride at various concentrations. Viability was measured by flow cytometry with the intercalating fluorescent probe propidium iodide, and the analysis was supplemented by electron microscopy. The highest resistance to osmotic stress was confirmed in the mutant strain with a deletion in the PHA synthase gene. At the same time, flow cytometry revealed a subpopulation of cells with significantly reduced internal complexity, the number of which increased with increasing stress levels. Initially, this subpopulation was detected only in the phaC mutant strain. However, upon repeating the experiment, similar cells were also observed in the other studied strains, including the wild-type strain.

Obhajoba proběhla podle následujícího schématu: prezentace studentky-vyjádření vedoucí/ho-oponentský posudek-reakce na posudek-diskuse s komisí. Studentka přednesla výborný výtah výsledků své bakalářské práce, řádně zodpověděla všechny dotazy oponentské i členů komise, pohotově reagovala na připomínky. V diskusi tak studentka prokázala výbornou schopnost orientace v teoretických i praktických základech problematiky bakalářské práce. Komise zhodnotila její bakalářskou práci celkově jako výbornou. Pekař: Jak moc statisticky významné jsou hodnoty na desetinných místech? Obruča: Jak si vysvětlujete hypotézu, že prázdné buňky brání ty živé? Kozáková: A proč se používá 600 nm? Enev: Není lepší používat pro optickou hustotu pojem turbiditu? Dzik: Jaké je měřítko snímků z elektronového mikroskopu?
