This bachelor’s thesis focuses on the optimization of the preparation of plasma-treated liquids (PTL) and their decontamination effects against the bacterium Cutibacterium acnes. PTL was prepared using a plasma system with a high-frequency alternating voltage source under simultaneous air bubbling. The study investigated the influence of cooling and treatment duration on PTL properties. Characterization was performed by measuring pH, conductivity, and the concentrations of reactive species. The results showed that preparation with cooling leads to PTL with higher concentrations of reactive species, while pH and conductivity did not differ significantly compared to preparation without cooling. With increasing treatment time, the concentrations of all monitored reactive species increased. The decontamination effect was tested on Cutibacterium acnes, and it was found that PTL treated for 10 minutes exhibited stronger antimicrobial effects than PTL treated for 3 minutes. For PTL10min, complete decontamination was achieved after 8 minutes of exposure, whereas PTL3min did not achieve complete decontamination even after 45 minutes. The aging of the inhibition efficacy of PTL10min was also investigated. The results showed that with increasing time after preparation, the antimicrobial effect gradually decreased. However, PTL still retained significant activity even 7 hours after preparation. The obtained results showed that preparation with cooling using an ice bath is more advantageous and confirmed the decontamination efficacy of PTL, and thus its potential for biomedical applications.

Obhajoba proběhla podle následujícího schématu: prezentace studentky-vyjádření vedoucí/ho-oponentský posudek-reakce na posudek-diskuse s komisí. Studentka přednesla výborný výtah výsledků své bakalářské práce, řádně zodpověděla všechny dotazy oponentské i členů komise, pohotově reagovala na připomínky. V diskusi tak studentka prokázala výbornou schopnost orientace v teoretických i praktických základech problematiky bakalářské práce. Komise zhodnotila její bakalářskou práci celkově jako výbornou. Pekař: Čím se ředily vzorky? Má NaCl vliv na produkci reaktivních částic? Obruča: Jak se stanovovaly různé časy expozice? Enev: Už se PTL testuje i na živých psech?
