This bachelor's thesis deals with the preparation of a plasma-treated hydrogel (PTH) based on sodium alginate from a previously plasma-treated saline solution (PTL) and the subsequent study of the release of reactive oxygen and nitrogen species (RONS), while various parameters that may affect RONS release were investigated, specifically the influence of the prepared PTH size, medium, storage temperature, and plasma treatment duration. The plasma-treated hydrogel was prepared by ionic crosslinking of sodium alginate with calcium chloride in the form of hydrogel beads, which were prepared by a custom (manual) method and using an encapsulator. The results indicate that all monitored parameters influenced RONS release. The highest concentrations were observed for hydrogels prepared from PTL treated for 10 minutes, stored in saline solution and at reduced temperature, while the effect of PTH size became apparent only for larger differences in PTH size. This bachelor's thesis also included an application experiment focused on long-term RONS release. The results indicate that RONS are able to persist in saline solution for a long period after being released from PTH, while chemical transformations between individual RONS may simultaneously occur. This ability of long-term persistence of RONS in the surrounding medium suggests the potential of plasma-treated hydrogels for future use in medical and decontamination applications.
