The bachelor´s thesis deals with the possibilities of using colloidal transport systems to increase the solubility and stability of poorly soluble active substances, directly following up on a previous study of the micellar properties of bile salts. Curcumin was used as a model drug. Within the experimental part, its photostability in solution and in the solid phase was evaluated first. The main objective was to assess the solubilization properties of bile salts, specifically sodium cholate, sodium deoxycholate, and sodium taurodeoxycholate, and a commercial lipid emulsion, Intralipid. The stability results confirmed the high photolability of curcumin in solution and its resistance in the crystalline form. Filtration through a 0.45 µm membrane was unreliable due to the passage of fine microparticles, while intensive centrifugation led to the undesirable sedimentation of even the solubilized fraction of curcumin. For Intralipid, a high solubilization capacity was visually confirmed; however, the UV-Vis determination was limited by light scattering on the lipid droplets. Due to these non-standard results, the dynamic light scattering (DLS) method was used at the end to characterize the particle size of curcumin in water. The DLS method confirmed that without modification, curcumin forms unstable macro-aggregates in water, whereas filtration leads to the retention of nanoparticles with a maximum around 100 nm. The obtained knowledge about the limitations of laboratory techniques and the behaviour of colloidal phases could contribute to the further development of non-polar drug carriers inspired by the mechanism of lipid digestion.

Studentka odpověděla na všechny doplňující otázky členů komise, které byly v průběhu diskuse k dané problematice vzneseny. V diskusi studentka prokázala výbornou orientaci v dané problematice. Po diskusi následovalo hodnocení závěrečné práce. Studentka prokázala nejen výborné odborné znalosti, ale i schopnost samostatné prezentace dosažených výsledků.
