This bachelor’s thesis focuses on the study of nucleic acid encapsulation into liposomal and chitosan-modified particles, as well as the monitoring of the effects of various temperature conditions on their stability during storage. Nucleic acids have a wide range of applications in medicine, from oncology and virology to genome editing. However, their use is significantly limited by their low stability under physiological conditions. To improve stability, prolong the release of the active substance, and enable targeted delivery, carrier systems are employed. As part of the optimization of nanoparticle preparation with nucleic acids, plasmid deoxyribonucleic acid of varying sizes and conformations was isolated in the experimental section and subsequently encapsulated into liposomal particles, some of which were coated with chitosan. The prepared formulations were characterized using dynamic light scattering to determine particle size and polydispersity, while the zeta potential was also measured as an indicator of stability of prepared formulations. The efficiency of encapsulation was determined using horizontal gel electrophoresis. Furthermore, changes in particle properties over time were monitored during both short-term and long-term storage at temperatures of 4 °C, 25 °C, and 37 °C. The amount of released pDNA was also analysed over time using horizontal gel electrophoresis. The prepared liposomes exhibited suitable properties for potential therapeutic applications and good temporal stability, whereas chitosan-modified particles did not reach these parameters. It was also found that sonication may have a destructive effect on pDNA and therefore may not be suitable for the preparation of these nanoparticles.

Izoloval jste plazmidovou DNA? Docházelo k transformaci? Jakou měl izolovaný plazmid velikost? Nedošlo při fragmentaci ke vzniku lepivých konců? Jakou metodou byly připravovány částice? Měřili jste koncentraci DNA před a po enkapsulaci? Jak? doc. Ing. Filip Mravec, Ph.D. Doporučení k úpravě spektrofotometrické techniky, aby nedocházelo k interferencím. prof. Ing. Jak funguje hydratace na tenké vrstvě? doc. Ing. Pavel Diviš, Ph.D. Na elektroforegramu v diskuzi oponentních otázek je zvláštní útvar kanálkovitého tvaru, čím mohl být způsoben? Student odpověděl na všechny doplňující otázky členů komise, které byly v průběhu diskuse k dané problematice vzneseny. V diskusi student prokázal výbornou orientaci v dané problematice. Po diskusi následovalo hodnocení závěrečné práce. Student prokázal nejen výborné odborné znalosti, ale i schopnost samostatné prezentace dosažených výsledků.
