This bachelor’s thesis focuses on the analysis of synthetic dyes in beverages available on the Czech and European markets using high-performance liquid chromatography with diode-array detection (HPLC–DAD). The work included the optimization of chromatographic conditions, the selection of a suitable mobile phase, and the determination of detection wavelengths based on the UV/VIS absorption spectra of the analyzed dyes. The analyses were performed on a Kinetex EVO C18 column (150 × 4.6 mm; 2.6 μm; 100 Å) using a mobile phase consisting of acetonitrile and 20 mM ammonium acetate adjusted to pH 5. Gradient elution was applied at a flow rate of 1 mL/min. The optimized HPLC–DAD method was used to identify and quantify selected synthetic dyes: tartrazine (E 102), Sunset Yellow FCF (E 110), Ponceau 4R (E 124), Allura Red AC (E 129), and Brilliant Blue FCF (E 133). The results were compared with the applicable EU legislative limits for the relevant beverage categories. In most of the analyzed samples, the determined concentrations were below the corresponding maximum limits. In one sample, the concentration of Sunset Yellow FCF was found to slightly exceed the limit value; however, this result should be interpreted with regard to measurement uncertainty and the method of legislative assessment for the given product type. Statistical evaluation of retention times confirmed good reproducibility of the method for most of the monitored dyes. The results provide an overview of the occurrence of selected synthetic dyes in the analyzed beverages and confirm the applicability of the HPLC–DAD method for their qualitative and quantitative determination.
