The combination of holographic microscopy with a low-coherence light source and confocal fluorescence microscopy enhances the capabilities of advanced live-cell imaging and real-time monitoring of dynamic biological processes. A newly developed confocal fluorescence module based on a DMD chip has been designed as an add-on unit for the hiQPI microscope. During the initial testing phase, experiments were conducted to verify the module’s basic functionality, focusing on fluorescence detection, depth selectivity, and lateral resolution. The results confirmed the system’s operational capability but revealed limited light efficiency. This limitation can be addressed through the use of a more powerful illumination source, a more sensitive detection system, or a more efficient scanning method. Based on the experimentally validated parameters, a mechanical integration concept was developed for incorporation into the hiQPI system. Optical simulations in Zemax were performed to optimize the distance between the tube lens and the sample in order to avoid beam clipping. The project also includes a detailed mechanical design in SolidWorks, featuring precision adjustment mechanisms compatible with both the hiQPI microscope and the confocal module. To verify the design, the components were fabricated using 3D printing and integrated into the mechanical assembly.
