This master's thesis presents a theoretical and experimental investigation of a novel application of geometric-phase holographic microscopy, enabling the measurement of the orientation of anisotropic structures in materials with spatially varying uniaxial birefringence. The introductory part provides a review of optical microscopy methods used for imaging both isotropic and anisotropic samples. The central part of the thesis focuses on the description of polarization states using Jones formalism and the optical components employed for their control. The acquired theoretical insights are then applied in the description of the imaging process, which analyzes the changes in the polarization state of light as it passes through an anisotropic sample, the microscope, and its geometric-phase holographic module. Here, the anisotropy orientation reconstruction process is also explained and numerical simulations are used to verify the theoretical concepts. The last part summarizes conducted experiments including a description of the microscope equipped with a geometric-phase holographic module and its application in the analysis of optically anisotropic samples. The obtained measurement results are compared with those from conventional polarization microscopy.
