This diploma thesis deals with the innovation of collagen vascular graft manufacturing technology in cooperation with VUP Medical a.s. The aim of this work was to design and experimentally verify a suitable method for collagen crosslinking that could be used in the current manufacturing process of vascular prostheses. The experimental part investigates different approaches to collagen crosslinking. Based on the obtained results, a method of photochemical crosslinking using riboflavin and UV radiation was selected, and collagen thin films and vascular prostheses were prepared. The prepared samples were evaluated in terms of stability in aqueous environment and vascular graft leakage. The samples were also analyzed using FTIR spectroscopy, tensile testing and atomic force microscopy (AFM). The results showed that riboflavin improved the stability of collagen material in aqueous environment and reduced the leakage of laboratory-prepared vascular prostheses. There were also observed changes in organization of the collagen structure and in mechanical behavior of the materiál itself. The obtained results suggest that photochemical collagen crosslinking using riboflavin could be a promising approach for further development of collagen vascular graft manufacturing technology.

Obhajoba proběhla podle následujícího schématu: prezentace studentky-vyjádření vedoucí/ho-oponentský posudek-reakce na posudek-diskuse s komisí. Studentka přednesla výborný výtah výsledků své diplomové práce, řádně zodpověděla všechny dotazy oponentské i členů komise, pohotově reagovala na připomínky. V diskusi tak studentka prokázala výbornou schopnost orientace v teoretických i praktických základech problematiky diplomové práce. Komise zhodnotila její diplomovou práci celkově jako výbornou. Kozáková: Jak probíhalo ošetření UV u cévní náhrady? Dzik: Jak probíhalo síťování po chemické stránce?
