This diploma thesis deals with the assessment of the effect of plasma activated water (PAW) on the germination and growth of pea (Pisum sativum L.) in an environment contaminated with heavy metals, specifically cadmium and lead. The aim of the work was to determine whether the application of PAW can mitigate the toxic effects of these contaminants and thus, support the growth and development of the plant. In the experimental part, PAW was prepared from tap water using corona discharge and its physicochemical properties were subsequently analyzed. The experiment with plants was divided into the evaluation of early growth on filter paper and 14-day hydroponic cultivation in solutions. The solutions were divided into three levels of contamination: low (5 mg/l), medium (50 mg/l) and high (100 mg/l). The development of morphological parameters, chlorophyll fluorescence parameters and element accumulation in plants was monitored using atomic absorption spectroscopy (AAS). The results show a stimulating effect of PAW at lower and medium concentrations of cadmium (5 and 50 mg/l), reducing the amount of toxic cadmium accumulation and promoting the uptake of essential elements such as magnesium and zinc. In the case of lead contamination and high cadmium contamination (100 mg/l), the stimulating effect of PAW was not confirmed. The combination of oxidative stress generated by PAW and heavy metal led to growth inhibition. The work thus confirms the stimulating potential of PAW for mitigating lower abiotic stress in agricultural applications, however, under extreme environmental stress, its application may have phytotoxic effects.

Obhajoba diplomové práce proběhla podle následujícího schématu: prezentace studenta-vyjádření vedoucí/ho-oponentský posudek-reakce na posudek-diskuse s komisí. Student přednesl výborný výtah výsledků své práce, řádně zodpověděl všechny dotazy oponentské i členů komise, pohotově reagoval na připomínky. V diskusi tak student prokázal výbornou schopnost orientace v teoretických i praktických základech problematiky práce. Komise zhodnotila jeho práci celkově jako výbornou. Veselý: Co je to konduktivita? Jančovičová: Nevadilo rostlinám nízké pH? Lehocký: Jaká je nejčastěji používaná voda v zemědělství?
