Simply method of DNA isolation from bacterial cells of genus Lactobacillus

Identifikace bakteriální DNA se skládá z několika kroků: příprava DNA v kvalitě vhodné pro polymerázovou řetězovou reakci (PCR), která zahrnuje lyzi buněk, extrakci a purifikaci DNA a následuje vlastní identifikace bakteriálního kmene pomocí PCR a jiných molekulárních metod. Pomocí roztoků pracích prášku obsahujících enzymy byly připraveny hrubé lyzáty buněk Lactobacillus plantarum CCM 7039, Lactobacillus fermentum CCM 7192, Lactobacillus rhamnosus CCM 1825, Lactobacillus casei subsp. casei CCM 7088. Z hrubých lyzátů buněk byla izolována DNA za použití metody vysolování. Kvalita eluované DNA a přítomnost cílové DNA byla ověřena metodou PCR, ve které byly použity specifické primery pro rod Lactobacillus LBLMA 1 a R16-1.

Identification of bacterial DNA consists of several steps: preparation of DNA with a quality suitable for polymerase chain reaction (PCR), which includes cell lysis, extraction and purification of DNA, followed by the identification of the bacterial strain by PCR and other molecular methods. Crude cell lysates of Lactobacillus plantarum CCM 7039, Lactobacillus fermentum CCM 7192, Lactobacillus rhamnosus CCM 1825, Lactobacillus casei subsp. Casei CCM 7088 were prepared with a washing powder solution containing enzymes. The DNA was isolated from the crude lysates using NaCl solution. Quality of the eluted DNA and the presence of the target DNA was verified by the PCR with specific primers for the genus Lactobacillus LBLMA 1 and R16-1.
