Original Abstract

A regenerative screening test is presented. Live cancer cell line HT1080 was exposed to selected drugs with a presumed antimigration activity that implies anti-metastatic potential. Four such migrastatic - niclosamide (NICL, 1 μM), belumosudil (BEL, 1 μM), midostaurin (MID, 1 μM) and vincristine (VIN, 100 nM) as the positive control were tested. The 20 hours - timelapse method with an objective lens of 10× 0.3 was chosen for a gradual regeneration test 3-times after gaps w/s treatment achieving thus the long-term monitoring of tumor cell activity. Holographic incoherent-light-source quantitative phase imaging (hiQPI) was used for this purpose. This approach allows for the most reliable and accurate automatic cell segmentation and monitors morphological and positional changes over time. This system is based on an off-axis setting with an incoherent light source. Therefore a clear high quality images without speckles and parasitic interferences can be achieved. The goal was to ensure a anti-metastatic effect that would be a compromise between reduced motility and increased cytotoxicity. All three migrastatics had a successful dampening effect on cell motility. At the same time, the cells were able to regenerate and restore their number after the substance was washed out. However, Niclosamide and Midostaurin achieved the best effect. This suppressive effect is also supported by statistics.
