Original Abstract

Taxonomy and phylogeny of bacterial species was always a very complex task, because bacteria do not have as many morphological signs as higher eukaryotes do. It gives rise to vast genotype-based techniques for their taxonomic relationships reassessment. While the former techniques, e.g. biotyping, chromosomal DNA fingerprinting or DNA-DNA hybridization, were carried out mostly in a laboratory, the latter approaches rely on in silico comparison of DNA sequences. No sooner was utilization of these techniques possible than the genome sequencing became widely available at reasonable costs. Even first experiments comparing only 16S rRNA genes lead to the reclassification of several solvent-producing strains, e.g. reclassification of C. acetobutylicum NCIMB 8052 as later C. beijerinckii NCIMB 8052. Unfortunately, the resolution of 16S rRNA gene itself was proven as insufficient to provide reliable classification of species or even strains. On the hand, recent advances in whole genome sequencing allow utilization of completely new in silico techniques on a genome-wide scale. We present genome comparisons of C. pasteurianum NRRL B-598, whose complete genome sequence we assembled last year, with several selected Clostridia using average nucleotide identity (ANI), digital DNA-DNA hybridization (dDDH) or several kinds of multilocus phylogenetic analyses. The results show that new kinds of genome-wide analyses are possible to bring other reclassifications among industrial solvent-producing Clostridia.
