In recent years, Laser Induced Breakdown Spectroscopy (LIBS) has become a promising technique for fast elemental analysis of biological tissues. However, optimizing laser parameters - in particular, the choice of wavelength - is crucial to increase analytical precision. This study presents a comparative analysis of three laser wavelengths (266 nm, 532 nm, and 1,064 nm) for LIBS-based analysis of soft biological tissues. Mouse liver tissues, embedded in paraffin blocks to maintain homogeneity and size consistency, served as the biological model. Key parameters, including laser energy (mJ), gate delay (GD), and atmospheric conditions (air vs. argon purge), were systematically optimized to evaluate their influence on signal quality and spatial resolution. Crater morphology using profilometer and irradiance values (GW/cm²) was also assessed to determine their impact on analytical performance. The results were visualized using heat maps, illustrating the relationship between irradiance, gate delay, and signal acquisition efficiency. Analysis revealed that although all wavelengths provided usable analytical outputs, one single wavelength outperformed the others in terms of higher spectral stability and analytical accuracy. In addition, the smaller crater sizes observed under optimized conditions at this wavelength suggest an advantage for high resolution, potentially used for histopathological imaging applications. This work highlights the critical role of laser parameter optimization in advancing LIBS for soft tissue characterization, with implications for biomedical research and clinical diagnostics.
